Pre-formed Fibrils (PFFs) Recombinant Proteins for Tau and SNCA

Alzheimer's disease (AD) and Parkinson's disease (PD) are the most common neurodegenerative disorders, characterized by the progressive accumulation and aggregation of specific proteins in the brain. In AD, hyperphosphorylated Tau protein forms neurofibrillary tangles, while in PD, α-synuclein (SNCA) forms Lewy bodies. These pathological protein aggregates not only serve as key disease markers but also actively propagate across neural networks, driving disease progression. Therefore, Tau PFFs and SNCA PFFs are essential tools for understanding disease mechanisms, developing diagnostic biomarkers, and screening potential therapeutics.

BrainVTA can provide high-quality Tau PFFs and SNCA PFFs with multiple variants for small or large scale research applications.

Product Specific Information

• Molecular Weight: Varies by variant (e.g., SNCA: ~14 kDa; Tau: ~40-50 kDa)
• Animal Origin Free: Yes
• Protein Length: Full-length or truncated variants as indicated
• Purity: ≥90% by SDS-PAGE
• Physical Characterization: Fiber formation validated by Transmission Electron Microscopy (TEM).
• Biological Activity: Validated in cell and animal models for seeding and propagation activity.

Testing Data

SDS-PAGE Analysis
The purity of each PFF product was assessed by SDS-PAGE. All variants showed a single predominant band at the expected molecular weight with purity ≥90%.

Transmission Electron Microscopy (TEM)
The formation of typical amyloid fibrils was confirmed by negative staining TEM. All PFF products exhibited characteristic long, unbranched fibrillar structures with diameters consistent with reported amyloid fibrils.

Applications

• Mechanistic studies of protein aggregation and prion-like propagation
• Construction of cell, animal, or organoid pathology models
• Anti-aggregation drug screening
• Comparative analysis of pathogenic properties and transmission differences among strains (e.g., PD, MSA, AD, PSP, CBD)
